    sammendrag:
Clostridium difficile infection (CDI) is a disease with symptoms ranging from mild diarrhea to severe life-threatening inflammation of the colon, generally in connection with a severe disturbance of the composition of intestinal microbiota. In the US, CDI is a very common disease, affecting approximately 500,000 patients annually, and of these around 30,000 will die from the disease. Live Biotherapeutic Products (LBP), are new alternative types of therapeutics, intended for the prevention of recurrence of CDI, by restoring the patients’ microbiota to a healthier condition. In this project, Genetic Analysis expertise in the microbiota field will be utilized to develop an easy-to-use, in vitro diagnostic test, by gut bacteria profile recognition, technical and clinical validation, and SW algorithm development. This will provide actionable results to allow for informed decisions regarding disease management and treatment and can be easily accessed by healthcare professionals.

Genetic Analysis is a Norwegian biotech company with a dedicated team of scientists with expertise within molecular biology, clinical research and bioinformatics/biostatistics. The company has previously CE-marked and launched the GA-map Dysbiosis test. The test is offered as an IVD version to laboratories in Europe and as "research use only" in the US. Genetic analysis has a production unit producing reagent kits for the Dysbiosis Test as well as an in-house service laboratory facility. The majority of the scientific staff is however working in a dedicated Research and Development team that constantly improves and develops new products for analysis of the human gut microbiota. This project will build on GAs expertise in human microbiota testing. In addition to developing new technological solutions for microbiota testing, GA will in this project explore further the function of viruses in the human gut as well as gaining a better understanding of the interplay between all microorganisms in the human gut (bacteria, viruses and parasites). This will be done by developing and improving novel technologies and methodologies for microbiota assays. GA will also aim to gain novel insight into the causal relationship between microbiota composition and clinical conditions and indications with the ultimate goal of enabling physicians to provide personalized and targeted treatment to patients.

In addition to developing new and improved technological solutions for microbiota testing, GA will also aim to gain novel insight into the causal relationship between microbiota composition and clinical conditions and indications with the ultimate goal of enabling physicians to provide personalized and targeted treatment to patients.

Several diseases have been linked to changes in the intestinal microbiota composition and function, ranging from gastrointestinal to neurological and autoimmune diseases. Playing a central role in human health, microbiome is today one of the most published topics in gut medical scientific journals. Testing the microbiome is a crucial step toward advancing knowledge in the area, but the lack of easy standardized tests to determine microbiome composition makes it difficult to assess and compare results in research studies. Microbiota also varies across geographies and ethnicities, and research is needed to understand differences in microbiota across populations. Genetic Analysis is a Norwegian science-based diagnostic company and pioneer in the human microbiota field. Our GA-map® platform is based on a pre-determined target multiplex approach specialized for simultaneous analysis of many bacteria in one reaction. The test results are generated by utilizing the patented and clinically validated cutting edge GA-map® software algorithm. This enables immediate results without the need for further bioinformatics work. Leveraging our experience from successful development and promising growth in European and US markets, we now aim to develop innovative diagnostic solutions for the rapidly growing human microbiota market in China. We have signed a Laboratory Developed Test (LDT) agreement with Thalys Medical Technology Group Corporation, and this project marks the beginning of our collaboration, with necessary steps to prepare for the LDT. This includes establishing a reference lab with the GA technology platform and a healthy reference microbiota profile for the Chinese population. Through this project we will build service opportunities and capabilities essential for Thalys to develop and distribute tests based on our GA-map® technology in China. We will also gain new knowledge about healthy microbiota in the Chinese population, which will be key for further research.

As of today, there are no solutions able to predict if an individual ulcerative colitis (UC) patient will suffer from mild or severe disease course and finding the appropriate treatment scheme is therefore still an unmet medical need. The intestinal microbiota represents a dynamic environment in which the relative balance of the composition of pro- and anti-inflammatory bacteria species is known to be highly relevant. Mapping of the intestinal microbiota can therefore provide a unique insight to the mechanisms predicting disease course of ulcerative colitis patients, to find the most effective treatment scheme. In this project, Genetic Analysis will develop an easy to use, in vitro diagnostic test that predicts disease course and treatment escalation requirements in patients with ulcerative colitis. Through gut bacteria profile recognition, the test will provide actionable results to allow for informed decisions regarding disease management and treatment. Importantly, the intervention can be used by any molecular diagnostics laboratory, and will therefore be easily accessed by the health care. The development and validation of the test will be done in collaboration with highly experienced clinical partners. This project will build on GAs expertise in human microbiota testing and the overall aim is to provide novel insight into the causal relationship between microbiota composition and clinical conditions and indications. Ultimately this will enable physicians to provide personalized and targeted treatment to patients.

There is a need for replacing the explorative hypothesis generating next generation sequencing techniques with techniques that only target microorganisms with diagnostic value. GA will develop a novel technology called Liquid Array Detection that allows detection of complex bacterial communities in a single-tube multiplex reaction, which is both simple and cheap. LAD detection is achieved by combining different channels of fluorescence detection and multiple melting profiles per channel. This project will build on GAs expertise in human microbiota testing and the overall aim is to provide novel insight into the causal relationship between microbiota composition and clinical conditions and indications. Ultimately this will enable physicians to provide personalized and targeted treatment to patients.

Genetic Analysis is a Norwegian biotech company with a dedicated team of scientists with expertise within molecular biology, clinical research and bioinformatics/biostatistics, in particular within the microbiome field. The company has previously CE-marked and launched the GA-map® Dysbiosis test, a test intended for gut microbiota characterizations. The test is offered as an IVD version to laboratories in Europe and as "research use only" in the US. Clostridium difficile infection (CDI) is a disease with signs and symptoms ranging from mild diarrhea to severe life-threatening inflammation of the colon, generally in connection with a severe disturbance of the composition of intestinal microbiota. In the US Clostridium difficile infection is a very common diseases, affecting approximately 500,000 patients annually, and of these around 30,000 will die from the disease. Live Biotherapeutic Products (LBP), is a new alternative kind of therapeutics, intended for the prevention of recurrence of CDI, by restoring the patients’ microbiota to a healthier condition. An important part in the treatment and restoration process is to identify the need and the effect of the treatment by measuring the baseline status of the microbiota, follow the changes in the microbiota during treatment until finally achieve restoration to a healthier microbiota status. In this project, Genetic Analysis will develop an easy to use, in vitro diagnostic test that through gut bacteria profile recognition will provide actionable results to allow for informed decisions regarding disease management and treatment. Importantly, the intervention can be used by any molecular diagnostics laboratory, and will therefore be easily accessed by the health care.
